作 者:秦剑秋[1] 刘海燕[1] 林新勤[1] 范云燕[1] 黄莉[1] QIN Jian-qiu,LIU Hai-yan,LIN Xin-qin,FAN Yun-yan,HUANG Li(Nanning Province Center for Control and Prevention,Nanning 530011,China) 机构地区:[1]南宁市疾病预防控制中心,广西南宁530011 出 处:《中国卫生检验杂志》2017年第11期2679-2683,共5页Chinses Journal of Health Laboratory Technology 基 金:广西南宁市科技局科技攻关项目(201702127C) 摘 要:目的:了解南宁市A(H3N2)亚型流感病毒血凝素的抗原性和基因变异情况。措施:对分离到的A(H3N2)亚型流感病毒进行单向HI试验略论抗原性,并按不同时间、不同人群选取35株进行血凝素核苷酸全长序列测定,推导出其氨基酸序列,重点对血凝素重链区进行基因特性略论。结果:单向HI试验表明,2017年度南宁市分离到的A(H3N2)亚型流感病毒抗原性与2004年WHO推荐株A/福建/411/2002(H3)相比,后者HI效价比前者高4倍以上;与2017年国内代表株A/江西东湖/312/2017(H3)相比,HI效价无≥4倍异同。核苷酸和氨基酸序列略论结果表明,与A/福建/411/2002(H3)相比核苷酸和氨基酸同源性分别为97.99%和97.81%,平均替换数分别为19.8个和7.2个,涉及2个抗原决定簇和受体结合位点左侧壁;与A/江西东湖/312/2017(H3)相比核苷酸和氨基酸同源性均为98.40%,平均替换数分别为15.8个和5.2个,涉及2个抗原决定簇。结论:2017年南宁市分离的A(H3N2)亚型流感病毒已发生抗原漂移。Objective:To study the antigenic and genetic characteristics of influenza(H3N2) virus circulated in Nanning in 2017.Methods: Single-way hemagglutination inhibition(HI) tests were firstly used to analyze the antigenic characteristics of influenza(H3N2) virus isolated.The HA1 gene of 35 isolates collected from different time and crowd were sequenced and analyzed.Results: In line with results of single-way hemagglutination inhibition test,the variance of hemagglutination inhibition valence showed to be more than 4 times difference when the antigenicity of influenza(H3N2) virus compared with A/Fujian/411/2002(H3),less than 4 times difference when compared with A/Jianxidonghu/312/2017(H3).The results of nucleotide and amic acid sequence demonstrated that the similarities of nucleotide and amino acid sequence compared with A/Fujian/411/2002(H3) were 97.99% and 97.81% respectively,the average number of alternative were 19.8 and 7.2,which involved 2 antigen determinants and RBS;the similarities compared with A/Jianxidonghu/312/2017(H3) were both 98.40%,the average number of alternative were 15.8 and 5.2,which involved 2 antigen determinants.Conclusion: There was significant mutation of antigenicity and genetic characteristics of influenza(H3N2) virus in Nanning in 2017. 关 键 词:流感病毒 血凝素 抗原 基因 变异 ,阿拉伯语论文网站,阿拉伯语论文 |
