摘  要:目的略论泰国葛愈伤组织诱导率和次生代谢产物的作用,为优化其愈伤组织培养条件提供数据支撑。措施采用正交实验探讨6-苄氨基腺嘌呤(6-BA,A)、a-萘乙酸(NAA,B)、吲哚丁酸(IBA,C)浓度及培养部位(根、茎、叶,D)对愈伤组织诱导率及次生代谢产物(葛根素、大豆苷、染料木素)累积量的作用。结果不同条件处理的愈伤组织均有所生长,当对愈伤组织诱导率的作用顺序为D〉A〉C〉B,其极差分别为24.107,22.153,18.624及13.080,最优组合为A2B2C3D2;当对总次生代谢产物质量分数的作用顺序为A〉C〉D〉B,其极差分别为2.083,1.837,1.205及0.347,最优组合为A2B3C1D1。结论泰国葛愈伤组织诱导的最佳条件:6-BA 1.0 mg/L,IBA 0.5 mg/L,NAA 0.5 mg/L,茎部培养;黄酮类次生代谢产物积累的最佳条件:根部培养,6-BA 1.0 mg/L,IBA 0 mg/L,NAA 1.0 mg/L。实际生产中可根据此条件分步培养以获得最高收益。Objective To analyze the effects of the culture conditions on callus induction rate and secondary metabolites in the calli of Pueraria mirifica, to provide support for the optimization of the callus culture conditions. Methods The effect of the concentrations of 6-BA (A), NAA (B), IBA (C) and culture parts (root/stem/leaf, D) on callus induction rate and the cumulant of secondary metabolites (puerarin, daidzein, genistein) were studied by an orthogonal experiment. Results The callus treated by different conditions all grew. The descending order of the effects of the conditions on callus induction was D〉A〉C〉B, and the range was 24.107, 22.153, 18.624 and 13.080, respectively. The optimal combination of the conditions was A2B2C3D2. The descending order of the effects of the conditions on mass fraction of the total secondary metabolites was A〉C〉D〉B, and the range was 2.083, 1.837, 1.205 and 0.347, respectively. The optimal combination of the conditions was AEB3CID1. Conclusion The best conditions of callus induction in the calli ofPueraria mirifica are stem culture, 6-BA 1.0 mg/L, IBA 0.5 mg/L, NAA 0.5 mg/L. The best conditions of accumulation of flavonoids secondary metabolites are root culture, 6-BA 1.0 mg/L, IBA 0 mg/L, NAA 1.0 mg/L. Under these conditions, callus can be cultured step by step to obtain the highest yield in the practical production.
